In patient samples, TYROBP expression was elevated in macrophages and tracked with disease activity markers. In mice, deleting Tyrobp reduced dextran sulfate sodium (DSS)-induced colitis, preserved tight junctions, and strengthened the intestinal barrier. Mechanistically, loss of KARAP improved macrophage mitochondrial homeostasis through mechanistic target of rapamycin (mTOR)/mitophagy signaling, increased interleukin-10 (IL-10) production, and protected epithelial cells. The work positions KARAP as a candidate target for strategies that aim to restore mucosal repair rather than merely suppress inflammation. These findings link an immune adaptor to metabolic control of barrier protection.
